1) Yes by gently holding the stem of your plant between the web of your fingers in one hand while you invert the pot and gently tap the sides of the pot to relase the soil and then gently remove the pot to inspect the roots.
 
Q2 Unfortunately the question of trichome color alone is not the sole factor in flower ripeness. Floral structures that have "filled out" and become dense are also a factor. Different strains mature at different rates. The flushing question also does not entail possible use of a flushing solution of a chelated agent which cuts the flushing down into a matter of 3-7 days. Depending on if this was being used or not, I would start flushing at 5-10% amber in order to achieve 30% amber

Do you do this every time you check the plants, or do you look at the plant first and see symptoms of root bound and then check the plant?
 
For Q1 I thought you said without transplanting it from the pot lol
