Becca's B+, Blue Meanie, Purple Mystic From Spore Genetics & Blue Yeti LC Fun Run

All the literature that I've read always specified Rye grain as the grain of choice. Now there are vids on youtube of comparative grows where a bunch of different grains were tested with the same strain of mycelium. If I remember correctly, rye and corn won.

Cool, @Tokin Roll . I think you'll like them- they look like pudgy little snow men with brown hats. Hella potent too.

Rye & rye berry are same thing, rye berry is the whole grain and is more nutritious. You should get some good, fast growth with it.

Saturday was 15 days since dunking the Blue Yeti brick in water for 24 hours and still nada so I pretty much decided that was it, then yesterday when I went to dispose of the brick I found fluffy white around the bottom and in a few patches above, too. I can't tell if it is shroom mycelium or contam so now I've changed my mind again and am back in "watch & see" mode. More shrooms would be cool! Pics from yesterday:

The Blue Yeti AIO today from 3 angles. I think it's done. This puffy white stuff doesn't look rhizomorphic and no pins have formed. 19 days after dunk. 🍄

Prepped 8 rice bags by breaking up and squeezing the rice toward the bottom of the bag, forming the rice roughly into a cube shape and applied a dab of silicone caulk to each to act as a self healing injection port. I'll make the filtered air exchange holes in the tops of the bags just prior to inoculation after the silicone has cured thoroughly (tomorrow or Saturday).
Four will get 2.5mL each B+ liquid culture and the other 4 will be inoculated with 2.5mL each of Blue Meanie Cubes, also using liquid culture I prepared earlier.
For now I've left the Blue Yeti AIO to do it's thing until I need the SAB.

That's the very same reason I need to get these bags going, Tok. The expiration date is May, 2024, so now is the time! 🍄

The silicone SHIPs have all cured and bags labelled. This evening I prepared the filters for the rice bags. Each 1" square filter is 4 layers of 3M #1390 Micropore tape.

20240426_201535r.jpg

I'm using 4 layers after discovering 1 layer doesn't do much as it's far too porous. My test method was simply holding a length of the tape up to a light and looking at it with a 5X magnifier. It took four layers to get to where I couldn't see light through it, so 4 layers it is! I made 2 extras just in case.

I'm going to use my soldering iron to make the breather holes in the rice bags. Set to 375F it should be just hot enough to melt the plastic without burning it.

Immediately after I'll adhere a filter tape over the hole, spray the tape with 70% Iso, and microwave the bag for 90 seconds. The 70% Iso and steam from heating the bag will fully sterilize the filter, then after repeating 8 more times, it will be time to

Opps! I screwed up- The Micropore tape is 3M Part #1530, NOT the part number I posted yesterday. Thought I best correct that!

Still haven't inoculated but did some more prep. Moved the Blue Yeti brick from the SAB into a germination tray with a humidity dome as I think there's still a chance of another flush. We'll see!

I also wiped down the SAB, sprayed everywhere inside w/ 70% Iso, closed it back up with FAE ports open and left it to air out. Also got out two sterile 10cc syringes and two sterile 16Ga x 1.5" needles to fill with liquid cultures.
Got the liquid culture jars of B+ and Blue Meanie Cubes out of the refrigerator to warm up. I'll give them a spin on the mag stirrer to break up clumps before filling the syringes.

Thanks, @Tokin Roll! Mine seemed slow, but that may be because the All-In-One used millet instead of Rye Berries.

Today I filled the syringes and just finished punching holes, applying filters, hosing the filters with 70% Iso and microwaving each bag 90 to 100 seconds (until the bag inflates so I know the filter is passing steam}. The eight bags are resting and cooling now, and I'm reasonably certain I've maintained sterility. I'll inoculate in the SAB when the bag temperature drops below 80F.

Edited to add: The soldering iron, using a 3/16" screwdriver tip, worked very well for making the FAE vent holes, but had to use a setting of 475F on the Ben bags to get melting. Got nice round holes with well defined rim. Perfect!

Inoculation completed just a few minutes ago! 4 bags of B+ and 4 bags of BMC. Break up/shake up date should be around April 10 or 12. This time I laid the bags on their side so I can watch the bottom for signs of growth. Also improved my technique to get more inoculation points in the grain so they take off faster.

Day 4 at around 90 hours. They are all growing hella fast and you can spot rhizomorphic growth in several bags. Liquid cultures rock. :ganjamon: 🍄

The Blue Yeti brick looks like it's ready to set fruit any time. Still misting it and airing it several times a day.

The rice bags are doing so well I decided I better get some fresh substrate on order now. Going to give mushroomsupplies.com a try so I ordered a 5# bag of dung lovers casing mix. Not a sponsor here but their products look to be good quality.
The plan is to use half in each of 2 shoeboxes with 4 bags each nocced rice. They offered to sweeten the pot so I grabbed one of their All-In-One bags for another romp with the Blue Yeti for the fun at a great price point.

Day 5, around 120 hours since inoculation with liquid cultures. The bags look like they'll be ready to shake tomorrow or Tuesday, earlier than I thought. B+ and BMC growing very fast. Blue Yeti is just chilling again today.
